Semaglutide

Semaglutide is a synthetic 31-residue acylated peptide supplied by Nocap Research strictly as a research chemical for qualified laboratory research use only.

What is Semaglutide?

Semaglutide is a synthetic 31-amino-acid acylated peptide, an analogue of GLP-1(7-37), with CAS registry number 910463-68-2 and molecular formula C187H291N45O59. The literature classifies it as a GLP-1 receptor agonist based on receptor studies. Nocap Research supplies it as a lyophilized reference material for laboratory research only.

Chemical Identity

Semaglutide, also referenced by the development code NN9535, is a synthetic peptide composed of 31 amino acids and designed as an analogue of glucagon-like peptide-1 (7-37). It is catalogued under CAS registry number 910463-68-2 and is described by the molecular formula C187H291N45O59, with a molecular weight of approximately 4113.6 g/mol. In the pharmacological literature the molecule is classified as a GLP-1 receptor agonist. These are molecular characterizations and do not describe an effect in humans or animals. Nocap Research offers this compound solely as a reference material for in vitro laboratory investigation.

Structure and Modifications

Semaglutide differs from native GLP-1(7-37) in three places: an Aib residue at position 2 (position 8 in GLP-1 numbering), an arginine at position 28 (position 34), and an acylated lysine at position 20 (position 26) that carries a C18 fatty diacid attached through a gamma-glutamic acid and two oligo(ethylene glycol) linker units. The C-terminus is a free acid.

The medicinal chemistry behind these substitutions is described in the original discovery paper, and cryo-EM structures of the peptide bound to the GLP-1 receptor have since been published.

Physical Form, Handling, and Storage

The material is supplied as a lyophilized (freeze-dried) powder. As a laboratory reagent, lyophilized peptide is generally stored desiccated and protected from light, with long-term storage commonly at -20C or below and short-term working material at 2C to 8C. For laboratory work the powder is solubilized in a solvent chosen for the intended in vitro application. These notes describe general handling of the material as a research reagent. They are not instructions for use in humans or animals.
